  • Venetoclax


    Venetoclax has shown to be effective for the treatment of patients with chronic lymphocytic leukemia (CLL) with 17p depletion (deletion of the short arm of chromosome 17), in which have received at least one prior therapy. It is marketed under the brand name Venclexta® in the United Stated and Venclyxto in Europe. Venetoclax is an orally bioavailable, selective small molecule inhibitor of the anti-apoptotic protein Bcl-2, with potential antineoplastic activity. Venetoclax mimics BH3-only proteins, the native ligands of Bcl-2 and apoptosis activators by binding to the hydrophobic groove of Bcl-2 proteins, thereby repressing Bcl-2 activity resulting in restoration of intrinsic apoptotic processes in tumor cells. Bcl-2 protein is overexpressed in some cancers and plays an important role in the regulation of apoptosis. Its expression is associated with increased drug resistance and tumor cell survival; therefore, venetoclax activates processes in tumor cells that lead to their cell death.

  • Penciclovir


    Penciclovir is an antiviral topical cream indicated for the treatment of recurrent herpes labialis (cold sores) of the lips and face. Penciclovir is an acyclic guanine nucleoside analog that has demonstrated in vitro inhibitory activity against herpes simplex virus types 1 (HSV-1) and 2 (HSV-2), varicella-zoster virus (VZV) and Epstein-Barr virus. Penciclovir is phosphorylated via viral thymidine kinase to a monophosphate form, which is subsequently converted to penciclovir triphosphate by cellular kinases. Intracellular conversion to the triphosphate form is required for antiviral activity (2, 3, 4, 5). In vitro studies demonstrate that penciclovir triphosphate competitively inhibits HSV polymerase with deoxyguanosine triphosphate. Inhibition of DNA synthesis of virus-infected cells suppresses viral replication. In cells not infected with HSV, DNA synthesis is unaltered due to phosphorylation occurring in only within herpes infected cells. (2, 3, 4, 5, 6). Penciclovir is the active metabolite of the oral product famciclovir.
